Lipopolysaccharide (LPS) causes a systemic inflammatory response, which is characterized by high mortality and few signs, including metabolic disorders, respiratory acidosis, hypotension, and disruption of vital organs. Cytokine secretion and oxidative stress are the main features of this disease. Diagnosis and treatment of systemic inflammation (SI) remains a challenge. Korean Red Ginseng (RG) is one of the medicinal plants that show strong anti-oxidant effects. We aimed to study the protective effects of GM on systemic inflammatory response in mice and RAW 264.7 macrophages induced by LPS.

Mice treated with water and alcohol extract of RG for four weeks to prevent the inflammatory response. The results showed that the LPS toxin increased morbidity and mortality, and induced liver, kidney, and injured lungs manifested by biomarker deteriorated. Hypotension, hypomagnesemia, acidosis, and oxidative stress observed in septic rats. However, RG extract attenuated liver, kidney, and lung enzymes and metabolites in the group treated with anti-inflammatory and anti-oxidant. Furthermore, RG enhanced magnesium and blood pressure in the treatment group.

RAW 264.7 macrophages exposed to LPS interference in translocation and MAPK p65 / p38. However, the RG-pretreated cells was not significantly changed. In conclusion, RG reducing mortality and morbidity rates of treated mice – liver, kidney, and lung injury are protected in the treatment group through potentiation of anti-oxidant defense. RG is able to save on mitochondrial function, inhibits the activation of MAPK / p38 signaling and suppressing NF κ B p65-core cytoplasmic transport. Further studies are needed to examine the effect on chronic conditions in animal models and humans.

Docking protein-1 promotes macrophage inflammatory signaling in gastric cancer.

Docking protein-1 (DOK1) is a tumor suppressor that is often missing in the malignant cells, however, still have the ability to control the activities of immune receptors on cells adjacent stroma of the tumor microenvironment. Therefore, we hypothesized that overcome DOK1 may be useful for cancer immunotherapy. DOK1 DOK1 mRNA and protein expression were downregulated in tumor cells of gastric cancer patients (n = 249). In contrast, expression of the rise and set in the positive case of Epstein Barr Virus (EBV +) together with genes associated with macrophage biology and clinical immunotherapy targets such as programmed cell death-ligand-1 (PD-L1).

In macrophages derived from leukemia cell lines monocytic human, DOK1 (i) is induced by agonist anti-diabetic transcription factor peroxisome proliferator-activated receptor-gamma (PPAR?), (Ii) an increase in polarization towards the phenotype of inflammation, (iii) plus transcription factor -kB nuclear dependent pro-inflammatory cytokines and (iv) reduce the expression of PD-L1.
